TGFb Effects on Primary Human Tenon Fibroblasts in Regard to Ocular Wound Healing

Purpose: A major complication after trabeculectomy consists in the postoperative scarring with consecutive therapeutic failure. The growth factor transforming growth factor b (TGFb) plays a pivotal role in the process of wound healing – in the eye, TGFb2 appears to be the predominant isoform. This study focuses on the effect of TGFb isoforms on primary human tenon fibroblasts (HTF) as well as on the fibroblast cell line NIH 3T3 with special regard to ocular wound healing.
Method: Primary human tenon fibroblasts (HTF) were expanded in culture - their TGFb receptor status was assessed by RT-PCR and by immunohistochemistry. HTFs and NIH 3T3 cells were stimulated with different concentrations of TGFb isoforms and activin A for various time periods. Activation of the SMAD signalling pathway was determined by western blot using phosphospecific antibodies. Expression of SMAD7, a negative regulator of the SMAD signal transduction, was assessed by western blot.
