Transport Influences on the Endothelium of Corneal Grafts in Organ Culture

Purpose: Experiments were performed to evaluate to what extent low temperature and mechanical shock are tolerated by the endothelium of corneal grafts in organ culture.
Method: Forty corneal grafts from 20 donors not suitable for keratoplasty were kept in organ culture for 7 to 28 days at 34.5°C. After this period, the grafts were divided into 8 groups: (A1) Organ culture at 20°C for 12 hours, thereafter organ culture at 34.5°C for 7 days; (A2) Control for group A1, preserved at 34.5°C over the whole culture period; (B1) Organ culture at 10° C for 12 hours, thereafter organ culture at 34.5°C for 7 days; (B2) Control for group B1, preserved at 34.5°C over the whole culture period; (C1) Mechanical shock exerted 100 times (>100g), thereafter organ culture at 34.5°C for 7 days; (C2) Control for group C1, preserved at 34.5°C over the whole culture period without exerting mechanical shock; (D1) Organ culture at 10° C for 12 hours, thereafter mechanical shock exerted 100 times (>100g), thereafter organ culture at 34.5°C for 7 days; (D2) Control for group D1. Each group consisted of 10 grafts. Endothelial evaluation was performed via digital imaging immediately before lowering temperature or exerting mechanical shock, 24 hours thereafter and at the end
